    针对已有报道的丹参素冰片酯合成工艺中成本高、实验条件相对苛刻、原料利用率低等缺陷和不足,以3,4-二羟基苯甲醛为起始原料,利用文献方法,在合成β-(3,4-二羟基苯基)丙酮酸冰片酯的基础上,利用活性酵母细胞代谢过程中产生的辅酶,生物催化不对称合成了丹参素冰片酯。系统考察了葡萄糖的加入量、反应时间、pH及β-环糊精加入量等因素对面包酵母生物催化合成不对称丹参素冰片酯的影响,实验结果表明:在底物β-(3,4-二羟基苯基)丙酮酸冰片酯加入量为为0.5g时,其最佳反应条件为葡萄糖30g、β-环糊精1.5g、pH为7、反应时间24h,最佳实验条件下ee值高达84.9%,产物主要为S构型的丹参素冰片酯。该工艺反应条件温和、成本较低,是一条极具价值的丹参素冰片酯生产工艺。

    Abstract:

    In view of the disadvantages of high cost、hash experimental conditions and low utilization rate of raw materials in existing reports. In the paper, 3,4-dihydroxybenzaldehyde was used as raw material, refer to the literature method, and β-(3,4-dihydroxyphenyl) pyruvate borneol ester was synthesized at first, then under the bread’s yeast fermentation liquid and target product Danshensu borneol ester was synthesized. The influence of amount of glucose and β-cyclodextrin、reaction time 、pH were investigated in the paper. When β-(3,4-dihydroxyphenyl) pyruvate borneol ester was 0.5g , And the best conditions were as follow: amount of glucose was 30g , reaction time was 24h, pH was 7, amount of β-cyclodextrin was 1.5g. enantiomeric excess valve of the target product as high as 84.9%,and main configuration was S-DBZ . The reaction conditions of the process in this paper were mild and the cost was low, which was a very valuable process for the production of Danshensu borneol ester.
