金花葵花黄酮超声提取工艺优化及性能分析

    摘要:

    为提高金花葵花中黄酮的提取率,探索金花葵在化妆品领域的应用价值,以金花葵花为原料,采用超声辅助提取法提取金花葵花黄酮,通过单因素实验和正交实验优化提取工艺,评估最佳工艺条件下提取的金花葵花黄酮的抗氧化活性及安全性。结果表明,金花葵花黄酮最佳提取工艺为:超声功率300 W,乙醇体积分数80%,超声时间35 min,提取温度75 ℃,料液比(g∶mL)1∶15,金花葵花黄酮最高提取率为89.97%。质量浓度为1 g/L金花葵花黄酮对1,1-二苯基苦基苯肼(DPPH)自由基、羟基自由基和2,2-联氮-双(3-乙基-苯并噻唑-6-磺酸)二铵盐(ABTS)阳离子自由基的清除率最高,分别为94.81%、95.24%、82.8%;对三者的半数抑制质量浓度(IC50)分别为106、70.06和213.8 μg/mL。金花葵花黄酮质量浓度<0.4 mg/mL时,其对红细胞溶血率均<10%,属于轻度刺激评级;质量浓度为1 mg/mL的金花葵花黄酮对鸡胚绒毛尿囊膜无溶血。。

    Abstract:

    In order to improve the extraction rate of flavonoids from Hibiseu manihot L flower and explore its application in cosmetics, Hibiseu manihot L flower was used as raw material, ultrasonic-assisted extraction was used to extract flavonoids from Hibiseu manihot L flower. The extraction process was optimized by a single factor experiment and an orthogonal experiment, and the antioxidant activity and safety of flavonoids extracted from Hibiseu manihot L flower under the optimal process conditions were evaluated. The results showed that the optimum extraction process of flavonoids from Hibiseu manihot L flower was as follows: ultrasonic power 300 W, ethanol volume fraction 80%, ultrasonic time 35 min, extraction temperature 75 °C, solid-liquid ratio (g ∶ mL) 1:15. The maximum extraction rate of flavonoids from Hibiseu manihot L flower was 89.97%. 1 g/L flavonoids from Hibiseu manihot L flower had the highest scavenging rates of DPPH free radical and hydroxyl radical, which were 94.81% and 95.24%, respectively. The half maximal inhibitory concentration (IC50) values were 0.1060 and 0.07006 g/L, respectively. The flavonoids from Hibiseu manihot L flower with a mass fraction of 3.13% had the largest radical scavenging rate (85.61% ) on ABTS cation-free radical. When the concentration of flavonoids from Hibiseu manihot L flower was less than 0.4 mg/mL, the hemolysis rate of red blood cells was less than 10%, which belonged to the mild stimulation rating. Flavonoids from the Hibiseu manihot L flower at a concentration of 1 mg/mL had no hemolysis on the chick embryo chorioallantoic membrane.
