    本实验旨在通过建立离子液体辅助植物乳杆菌发酵体系,优化以刺五加为原材料异嗪皮啶的生物转化及提取工艺。对预处理离子液体阴离子成分、烷基链长度进行筛选,在此基础上研究了pH、液固比、发酵时间、发酵温度和离子液体浓度对异嗪皮啶产量的影响,进而进行响应面实验,最终确定在ILs[C4MIM]Br浓度0.57 M、发酵温度36.5℃、pH 6.0的条件下优化超声提取条件,超声功率 480W、液固比19mL/g、超声时间39 min时,得到异嗪皮啶产量为0.312mg/g,相较于未处理样品(0.051 mg/g)提高了6.12倍。该研究表明,植物乳杆菌具有较好的转化能力,在绿色催化剂离子液体的辅助下,能够显著提升异嗪皮啶的产量,具有较好的应用前景。

    Abstract:

    The aim of this experiment was to optimize the bioconversion and extraction process of Isofraxidin using Acanthopanax senticosus as raw material by establishing an ionic liquid-assisted Lactobacillus plantarum fermentation system. The pretreatment ionic liquid anion composition and alkyl chain length were screened, based on which the effects of pH, liquid-solid ratio, fermentation time, fermentation temperature and ionic liquid concentration on the yield of Isofraxidin were investigated, and then response surface experiments were carried out, and the optimized ultrasonic extraction conditions were finally determined at a concentration of ILs[C4MIM]Br of 0.57 M, a fermentation temperature of 36.5 °C and a pH of 6.0, with the following conditions. The yield of Isofraxidin of 0.312 mg/g was obtained at ultrasonic power of 480 W, liquid-solid ratio of 19 mL/g, and ultrasonic time of 39 min, which was 6.12-fold higher compared with the untreated sample (0.051 mg/g). This study demonstrated that Lactobacillus plantarum has a good transformation ability and can significantly enhance the yield of Isofraxidin with the assistance of ionic liquid, a green catalyst, which has a good application prospect.
